Serum Creatinine is an unusable chemical product derived from creatinine in human muscle. Creatinine is excreted from the body through the kidneys. Therefore, in the case of kidney failure, the level of creatinine in the blood starts to increase. The amount of creatinine in the blood is a reflection of kidney function. The blood urea test is less effective than the creatinine test.
Creatinine Test Kit:
(Common method) Jaffe Slot modified alkaline picrate calorimetric method is used. In this method, chemical components other than creatinine are not affected and it is considered to be a more effective method. The total test time with this method is 15 minutes while the total test time is 45 minutes with the second manual method using no kit.
Principles of Jeff Slot Method:
In this method for serum creatinine, picric acid reacts chemically with creatinine in an alkaline solution, resulting in a yellowish-reddish solution. The absorbance of this dye is recorded on the blue-green filter of the colorimeter or at a wavelength of 490 nanometers. This is called the first reading. Before taking the second reading, the color solution is acidified, causing the color of the color solution to become extremely cloudy due to creatinine.
The dye absorption capacity of this solution is again tested. The second reading is differentiated from the first reading because the second reading shows color due to chemical reactions with chemicals other than creatinine. The amount obtained after differentiation shows only creatinine.
Note:
This working reagent remains usable for only one day. Then prepare fresh reagent again.
Patient Sample:
This test requires 400 microliters of fresh patient serum or plasma.
